Submitter rationale: This SLC43A1 variant (rs201459634) is rare (<0.1%) in a large population dataset (gnomAD: 4/1610162 total alleles; 0.00025%; no homozygotes) and has not been reported in ClinVar nor the literature, to our knowledge. Three bioinformatic tools queried predict that this substitution would be tolerated, but these algorithms have low specificity, especially for predicting gain of function or dominant negative variants. The threonine residue at this position is evolutionarily conserved across many of the species assessed, although alanine is present at this position in birds. We consider the clinical significance of SLC43A1 c.130A>G to be uncertain at this time.
