NM_001277115.2(DNAH11):c.9210C>T (p.Thr3070=) was classified as Uncertain significance for Primary ciliary dyskinesia 7 by Johns Hopkins Genomics, Johns Hopkins University, citing ACMG Guidelines, 2015: This DNAH11 variant is absent from a large population dataset and has not been reported in ClinVar nor in the literature in individuals with primary ciliary dyskinesia 7, to our knowledge. Bioinformatic analysis predicts that this synonymous variant would not affect normal exon 56 splicing, although this has not been confirmed experimentally to our knowledge. Due to the lack of clinical and functional data supporting that this variant is deleterious, we consider the clinical significance of DNAH11 c.9210C>T to be uncertain at this time.
