NM_001395656.1(ROBO2):c.2953A>G (p.Thr985Ala) was classified as Uncertain significance for Vesicoureteral reflux 2 by Johns Hopkins Genomics, Johns Hopkins University, citing ACMG Guidelines, 2015: This ROBO2 missense variant (rs370723000 ) is rare (<0.1%) in a large population dataset (gnomADv4.0.0: 28/1613982 total alleles; 0.002%; no homozygotes). It has not been reported in ClinVar, nor the literature, to our knowledge. Of two bioinformatics tools queried, one predicts that the substitution would be damaging, while the other predicts that it would be tolerated, and the threonine residue at this position is evolutionarily conserved across all of the species assessed. We consider the clinical significance of c.2953A>G in ROBO2 to be uncertain at this time.
