NM_001387430.1(SH2B1):c.*17C>A was classified as Uncertain significance for obesity by Clinical Genomics Laboratory, Washington University in St. Louis, citing ACMG Guidelines, 2015: The SH2B1 c.*17C>A variant occurs in the 3' untranslated region (UTR) after the stop codon but before the polyadenylation signal. This variant, to our knowledge, has not been reported in the medical literature and is absent from the general population (gnomAD v.2.1.1), indicating it is not a common variant. A recently published computational program designed to predict the impact of 3'UTR variants (Romo L et al., PMID: 38237594) predicts that this variant may impact miRNA target sites, which may affect the stability of the mRNA. However, due to limited information, the clinical significance of this variant is uncertain.
