This variant lies in the FAT4 gene (transcript NM_001291303.3) at coding-DNA position 1168, where G is replaced by T; at the protein level this means replaces alanine at residue 390 with serine — a missense variant. Submitter rationale: A FAT4 c.1168G>T (p.Ala390Ser) variant was identified at a near heterozygous allelic fraction of 49.9%, a frequency which may be consistent with it being of germline origin. This variant, to our knowledge, has not been reported in the medical literature. It is only observed in 30/1,613,736 alleles in the general population (gnomAD v4.1.0), indicating it is not a common variant. Computational predictors suggest that the variant does not impact FAT4 function. Due to limited information, and based on ACMG/AMP guidelines for variant interpretation (Richards S et al., PMID: 25741868), the clinical significance of this variant is uncertain at this time.
