NM_001291303.3(FAT4):c.1364G>A (p.Arg455His) was classified as Uncertain significance for Van Maldergem syndrome 2; Hennekam lymphangiectasia-lymphedema syndrome 2 by Clinical Genomics Laboratory, Washington University in St. Louis, citing ACMG Guidelines, 2015: A FAT4 c.1364G>A (p.Arg455His) variant was identified at a heterozygous allelic fraction of 51.4%, a frequency which may be consistent with it being of germline origin. This variant, to our knowledge, has not been reported in the medical literature. It is only observed in 2/1,614,150 alleles in the general population (gnomAD v4.1.0), indicating it is not a common variant. Computational predictors are uncertain as to the impact of this variant on FAT4 function. Due to limited information, and based on ACMG/AMP guidelines for variant interpretation (Richards S et al., PMID: 25741868), the clinical significance of this variant is uncertain at this time.
