NM_004415.4(DSP):c.8014C>G (p.Gln2672Glu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q2672E variant (also known as c.8014C>G), located in coding exon 24 of the DSP gene, results from a C to G substitution at nucleotide position 8014. The glutamine at codon 2672 is replaced by glutamic acid, an amino acid with highly similar properties. This alteration has been reported in hypertrophic cardiomyopathy (HCM) cohorts; however, clinical details were limited (Robyns T et al. Eur J Hum Genet, 2017 Dec;25:1313-1323; Smith E et al. J Am Heart Assoc, 2022 May;11:e024501). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
