NM_001378454.1(ALMS1):c.9322G>A (p.Ala3108Thr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A3109T variant (also known as c.9325G>A), located in coding exon 10 of the ALMS1 gene, results from a G to A substitution at nucleotide position 9325. The alanine at codon 3109 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
