Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.3207G>T (p.Met1069Ile), citing Ambry Variant Classification Scheme 2023: The p.M1069I variant (also known as c.3207G>T), located in coding exon 20 of the ALK gene, results from a G to T substitution at nucleotide position 3207. The methionine at codon 1069 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
