Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.1953A>C (p.Lys651Asn), citing Ambry Variant Classification Scheme 2023: The p.K651N variant (also known as c.1953A>C), located in coding exon 11 of the ALK gene, results from an A to C substitution at nucleotide position 1953. The lysine at codon 651 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
