Pathogenic — the classification assigned by GeneDx to NM_013382.7(POMT2):c.462G>A (p.Trp154Ter), citing GeneDx Variant Classification (06012015): The W154X nonsense variant in the POMT2 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Although this variant has not been reported previously to our knowledge, other nonsense variants have been reported in the Human Gene Mutation Database in association with dystroglycanopathy (Stenson et al., 2014). Therefore, W154X is considered to be a pathogenic variant.
