This variant lies in the NONO gene (transcript NM_007363.5) at coding-DNA position 1009, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 337 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The R337X nonsense variant in the NONO gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. It is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret R337X as a pathogenic variant.
