Pathogenic — the classification assigned by GeneDx to NM_001195553.2(DCX):c.30_31del (p.Arg11_Asp12insTer), citing GeneDx Variant Classification (06012015): The c.30_31delAA variant in the DCX gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.30_31delAA variant is predicted to change amino acid Aspartic acid 12 into a premature Stop codon, denoted p.Asp12Ter. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.30_31delAA variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.30_31delAA as a pathogenic variant.
