Pathogenic — the classification assigned by GeneDx to NM_000094.4(COL7A1):c.2607_2608del (p.Leu870fs), citing GeneDx Variant Classification (06012015). This variant lies in the COL7A1 gene (transcript NM_000094.4) at coding-DNA position 2607 through coding-DNA position 2608, deleting 2 bases; at the protein level this means shifts the reading frame starting at leucine residue 870, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: The c.2607_2608delCC pathogenic variant in the COL7A1 gene causes a frameshift starting with codon Leucine 870, changes this amino acid to a Glycine residue and creates a premature Stop codon at position 33 of the new reading frame, denoted p.Leu870GlyfsX33. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Additionally, the c.2607_2608delCC variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Therefore, we interpret the c.2607_2608delCC variant as pathogenic.
