This variant lies in the KRT83 gene (transcript NM_002282.3) at coding-DNA position 745, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 249 with glutamine — a missense variant. Submitter rationale: The E249Q variant in the KRT83 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E249Q variant is observed in 1/10404 (0.01%) alleles from individuals of African background in the ExAC dataset (Lek et al., 2016). The E249Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position where amino acids with similar properties to Glutamic Acid are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret E249Q as a variant of uncertain significance.
