This variant lies in the TCAP gene (transcript NM_003673.4) at coding-DNA position 98, where G is replaced by A; at the protein level this means replaces arginine at residue 33 with glutamine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the TCAP gene. The R33Q variant has not been published as pathogenic or been reported as benign to our knowledge. Additionally, this variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R33Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. Furthermore, in silico analysis predicts this variant is probably damaging to the protein structure/function. Lastly, this substitution occurs at a position that is conserved in mammals, however, glutamine (Q) is the wild-type residue at this position in at least two non-mammalian species.
