NM_206933.4(USH2A):c.5776+2T>C was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.5776+2T>C variant in the USH2A gene has been reported previously in association with Usher syndrome type II (Bonnet et al., 2016). This splice site variant destroys the canonical splice donor site in intron 28. It is predicted to cause abnormal gene splicing, either leading to an abnormal message that is subject to nonsense-mediated mRNA decay, or to an abnormal protein product if the message is used for protein translation. The c.5776+2T>C variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). A canonical splice site variant in the same splice donor site in intron 28 (c.5776+1G>A) has also been reported in association with Usher syndrome (Sandberg et al., 2008; Jaijo et al., 2010; Wang et al., 2014; Glockle et al., 2014). We interpret c.5776+2T>C as a pathogenic variant.
