Uncertain significance — the classification assigned by GeneDx to NM_001330574.2(ZNF711):c.1873C>T (p.Arg625Cys), citing GeneDx Variant Classification (06012015). This variant lies in the ZNF711 gene (transcript NM_001330574.2) at coding-DNA position 1873, where C is replaced by T; at the protein level this means replaces arginine at residue 625 with cysteine — a missense variant. Submitter rationale: The R579C variant in the ZNF711 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R579C variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R579C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret R579C as a variant of uncertain significance.
