This variant lies in the GRIN2A gene (transcript NM_001134407.3) at the canonical splice acceptor site of the intron immediately before 18 bases upstream of the translation start (5' untranslated region), where A is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.-18-2A>T intronic alteration consists of a A to T substitution two nucleotides before exon 3 (coding exon 1) of the GRIN2A gene. Alterations that disrupt the canonical splice site are expected to result in aberrant splicing. The exact functional effect of this alteration is unknown.
