NM_013372.7(GREM1):c.91G>T (p.Gly31Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G31C variant (also known as c.91G>T), located in coding exon 1 of the GREM1 gene, results from a G to T substitution at nucleotide position 91. The glycine at codon 31 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
