Pathogenic — the classification assigned by GeneDx to NM_002890.3(RASA1):c.2798_2804del (p.Val933fs), citing GeneDx Variant Classification (06012015): The c.2798_2804delTGGCTAA variant in the RASA1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.2798_2804delTGGCTAA variant causes a frameshift starting with codon Valine 933, changes this amino acid to an Aspartic acid residue, and creates a premature Stop codon at position 6 of the new reading frame, denoted p.Val933AspfsX6. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.2798_2804delTGGCTAA variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.2798_2804delTGGCTAA as a pathogenic variant.
