Uncertain significance — the classification assigned by GeneDx to NM_001148.6(ANK2):c.10084T>A (p.Ser3362Thr), citing GeneDx Variant Classification (06012015). This variant lies in the ANK2 gene (transcript NM_001148.6) at coding-DNA position 10084, where T is replaced by A; at the protein level this means replaces serine at residue 3362 with threonine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the ANK2 gene. The S3362T variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Neverthelees, the S3362T variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. Additionally, this substitution occurs at a position that is not conserved, and threonine is tolerated at this position in several species. Furthermore, in silico analysis predicts this variant likely does not alter the protein structure/function.
