Uncertain significance — the classification assigned by GeneDx to NM_007289.4(MME):c.-10-1G>T, citing GeneDx Variant Classification Process June 2021. This variant lies in the MME gene (transcript NM_007289.4) at the canonical splice acceptor site of the intron immediately before 10 bases upstream of the translation start (5' untranslated region), where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: Not observed in large population cohorts (Lek et al., 2016); Has not been previously published as pathogenic or benign to our knowledge; In silico analysis, which includes splice predictors and evolutionary conservation, suggests this variant may impact gene splicing. In the absence of RNA/functional studies, the actual effect of this sequence change is unknown.
