Uncertain significance — the classification assigned by GeneDx to NM_005548.3(KARS1):c.904C>T (p.Leu302Phe), citing GeneDx Variant Classification (06012015). This variant lies in the KARS1 gene (transcript NM_005548.3) at coding-DNA position 904, where C is replaced by T; at the protein level this means replaces leucine at residue 302 with phenylalanine — a missense variant. Submitter rationale: The L330F variant in the KARS gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The L330F variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The L330F variant is a conservative amino acid substitution, which occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret L330F as a variant of uncertain significance.
