This variant lies in the AKAP9 gene (transcript NM_005751.5) at coding-DNA position 177 through coding-DNA position 263, duplicating 87 bases. Submitter rationale: The c.177_263dup87 variant (also known as p.Q59_L87dup), located in coding exon 2 of the AKAP9 gene, results from an in-frame duplication of 87 nucleotides at nucleotide positions 177 to 263. This results in the duplication of 29 extra residues (QSQCNEMYINSSQRVESTVIPESTIMRTL) between codons 59 and 87. This amino acid region is not well conserved in available vertebrate species. In addition, this variant is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
