NM_000314.8(PTEN):c.947del (p.Leu316fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.947delT variant in the PTEN gene has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This deletion causes a frameshift starting with codon Leucine 316, changes this amino acid to a Glutamine residue and creates a premature Stop codon at position 5 of the new reading frame, denoted p.Leu316GlnfsX5. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Based on currently available evidence we consider c.947delT to be pathogenic.
