Uncertain significance — the classification assigned by Ambry Genetics to NM_001130009.3(GEN1):c.125A>T (p.Lys42Ile), citing Ambry Variant Classification Scheme 2023: The p.K42I variant (also known as c.125A>T), located in coding exon 1 of the GEN1 gene, results from an A to T substitution at nucleotide position 125. The lysine at codon 42 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
