Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_032638.5(GATA2):c.52A>G (p.Asn18Asp), citing Ambry Variant Classification Scheme 2023: The p.N18D variant (also known as c.52A>G), located in coding exon 1 of the GATA2 gene, results from an A to G substitution at nucleotide position 52. The asparagine at codon 18 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
