NM_178014.4(TUBB):c.1230G>T (p.Glu410Asp) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The E410D variant in the TUBB gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E410D variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The E410D variant is a conservative amino acid substitution, which occurs at a position that is conserved across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret E410D as a likely pathogenic variant
