Uncertain significance — the classification assigned by Ambry Genetics to NM_024642.5(GALNT12):c.1031T>A (p.Phe344Tyr), citing Ambry Variant Classification Scheme 2023: The p.F344Y variant (also known as c.1031T>A), located in coding exon 5 of the GALNT12 gene, results from a T to A substitution at nucleotide position 1031. The phenylalanine at codon 344 is replaced by tyrosine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
