Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_138387.4(G6PC3):c.177G>C (p.Trp59Cys), citing Ambry Variant Classification Scheme 2023: The p.W59C variant (also known as c.177G>C), located in coding exon 1 of the G6PC3 gene, results from a G to C substitution at nucleotide position 177. The tryptophan at codon 59 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
