NM_015030.2(FRYL):c.492-2A>G was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.492-2A>G intronic variant results from an A to G substitution two nucleotides before coding exon 6 of the FRYL gene. Alterations that disrupt the canonical splice site are expected to result in aberrant splicing. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This nucleotide position is highly conserved in available vertebrate species. RNA studies have demonstrated that this alteration results in a splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). In silico splice site analysis predicts that this alteration will weaken the native splice acceptor site and will result in the creation or strengthening of a novel splice acceptor site. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
