Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_144997.7(FLCN):c.162G>C (p.Met54Ile), citing Ambry Variant Classification Scheme 2023: The p.M54I variant (also known as c.162G>C), located in coding exon 1 of the FLCN gene, results from a G to C substitution at nucleotide position 162. The methionine at codon 54 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
