Pathogenic — the classification assigned by GeneDx to NM_000088.4(COL1A1):c.458dup (p.Gly154fs), citing GeneDx Variant Classification (06012015): The c.458dupC pathogenic variant in the COL1A1 gene has been reported in association with osteogenesis imperfecta (Lindahl et al. 2015). This variant causes a frameshift starting with codon Glycine 154, changes this amino acid to a a Tryptophan residue and creates a premature Stop codon at position 15 of the new reading frame, denoted p.G154WfsX15. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay.
