Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_004629.2(FANCG):c.100G>A (p.Gly34Ser), citing Ambry Variant Classification Scheme 2023: The p.G34S variant (also known as c.100G>A), located in coding exon 2 of the FANCG gene, results from a G to A substitution at nucleotide position 100. The glycine at codon 34 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
