Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000135.4(FANCA):c.1350C>G (p.Asp450Glu), citing Ambry Variant Classification Scheme 2023: The p.D450E variant (also known as c.1350C>G), located in coding exon 14 of the FANCA gene, results from a C to G substitution at nucleotide position 1350. The aspartic acid at codon 450 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
