This sequence change replaces serine, which is neutral and polar, with glycine, which is neutral and non-polar, at codon 50 of the MYBPC3 protein (p.Ser50Gly). This variant is present in population databases (rs373164247, gnomAD 0.005%). This missense change has been observed in individual(s) with hypertrophic cardiomyopathy and dilated cardiomyopathy (PMID: 27532257, 37652022). ClinVar contains an entry for this variant (Variation ID: 42538). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The glycine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. (less)

p.Ser50Gly in exon 2 of MYBPC3: This variant is not expected to have clinical si gnificance because the serine (Ser) residue at position 50 is not conserved in mammals or evolutionarily distant species and >10 mammals carry a glycine (Gly) at this position. In addition, this change was predicted to be benign using a co mputational tool clinically validated by our laboratory. This tool's benign pred iction is estimated to be correct 89% of the time (Jordan 2011). This variant ha s also been identified in 3/43654 European chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadinstitute.org; dbSNP rs373164247). (less)
