Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_020207.7(ERCC6L2):c.724A>T (p.Arg242Trp), citing Ambry Variant Classification Scheme 2023: The p.R242W variant (also known as c.724A>T), located in coding exon 4 of the ERCC6L2 gene, results from an A to T substitution at nucleotide position 724. The arginine at codon 242 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
