This variant lies in the CASR gene (transcript NM_000388.4) at coding-DNA position 893, where C is replaced by T; at the protein level this means replaces alanine at residue 298 with valine — a missense variant. Submitter rationale: Variant summary: CASR c.893C>T (p.Ala298Val) results in a non-conservative amino acid change located in the Receptor, ligand binding region domain (IPR001828) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251232 control chromosomes. c.893C>T has been reported in the literature in individuals affected with features of Familial Hypocalciuric Hypercalcemia (example, Mouly_2020, Lasbleiz_2022). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 34714514, 32347971). Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
