This variant lies in the CACNA1C gene (transcript NM_000719.7) at coding-DNA position 3962, where C is replaced by T; at the protein level this means replaces serine at residue 1321 with phenylalanine — a missense variant. Submitter rationale: The S1321F variant in the CACNA1C gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S1321F variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The S1321F variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals and in silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret S1321F as a variant of uncertain significance.
