Pathogenic for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004415.4(DSP):c.4639dup (p.Gln1547fs), citing Ambry Variant Classification Scheme 2023: The c.4639dupC (p.Q1547Pfs*80) alteration, located in exon 23 (coding exon 23) of the DSP gene, consists of a duplication of C at position 4639, causing a translational frameshift with a predicted alternate stop codon after 80 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This variant was reported in individual(s) with features consistent DSP-related cardiomyopathy (Gasperetti, 2025). Based on the available evidence, this alteration is classified as pathogenic.
