NM_024422.6(DSC2):c.712G>T (p.Asp238Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D238Y variant (also known as c.712G>T), located in coding exon 6 of the DSC2 gene, results from a G to T substitution at nucleotide position 712. The aspartic acid at codon 238 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
