NM_206933.4(USH2A):c.2384G>A (p.Cys795Tyr) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the USH2A gene (transcript NM_206933.4) at coding-DNA position 2384, where G is replaced by A; at the protein level this means replaces cysteine at residue 795 with tyrosine — a missense variant. Submitter rationale: The C795Y variant in the USH2A gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The C795Y variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The C795Y variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret C795Y as a likely pathogenic variant.
