Uncertain significance — the classification assigned by GeneDx to NM_001148.6(ANK2):c.3399C>G (p.Asp1133Glu), citing GeneDx Variant Classification (06012015): The D1133E variant of uncertain significance in the ANK2 gene has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). However, the D1133E variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to aspartic acid are tolerated across species, and glutamic acid is the wild-type amino acid at this position in multiple species. Furthermore, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
