NM_001142864.4(PIEZO1):c.3940C>T (p.Leu1314Phe) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The L1314F variant in the PIEZO1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The L1314F variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The L1314F variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to Leucine are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret L1314F as a variant of uncertain significance.
