NM_000135.4(FANCA):c.3655G>A (p.Ala1219Thr) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The A1219T variant in the FANCA gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The A1219T variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The A1219T variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant likely does not alter the protein structure/function. We interpret A1219T as a variant of uncertain significance.
