NM_017636.4(TRPM4):c.3563G>T (p.Trp1188Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W1188L variant (also known as c.3563G>T), located in coding exon 24 of the TRPM4 gene, results from a G to T substitution at nucleotide position 3563. The tryptophan at codon 1188 is replaced by leucine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
