This variant lies in the TBX1 gene (transcript NM_001379200.1) at coding-DNA position 823, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 275 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E266X nonsense variant in the TBX1 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Although this variant has not been reported previously to our knowledge, we consider it to be pathogenic.
