Uncertain significance — the classification assigned by GeneDx to NM_139281.2(WDR36):c.5G>A (p.Cys2Tyr), citing GeneDx Variant Classification (06012015). This variant lies in the WDR36 gene (transcript NM_139281.2) at coding-DNA position 5, where G is replaced by A; at the protein level this means replaces cysteine at residue 2 with tyrosine — a missense variant. Submitter rationale: The C2Y variant in the WDR36 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The C2Y variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The C2Y variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret C2Y as a variant of uncertain significance.
