Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.2038G>T (p.Val680Phe), citing Ambry Variant Classification Scheme 2023: The p.V680F variant (also known as c.2038G>T), located in coding exon 11 of the DICER1 gene, results from a G to T substitution at nucleotide position 2038. The valine at codon 680 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
